محلول ترایزول (با برند تجاری YTzol pure RNA شرکت یکتا تجهیز آزما ) یک معرف آماده استفاده بر پایه فنول-گوانیدین است که به منظور جداسازی RNA با کیفیت بالا از انواع نمونه های بیولوژیکی طراحی شده است.
این معرف، یک محلول تک فازی از فنول، گوانیدین ایزوتیوسیانات و سایر اجزای اختصاصی است که جداسازی انواع RNA با اندازه های مولکولی بزرگ یا کوچک را تسهیل می کند.
محلول YTzol برای جداسازی چندین هدف مولکولی ساخته شده بنابراین این امکان را به شما می دهد تا علاوه برRNA ، DNA و پروتئین ها را نیز از یک نمونه واحد استخراج کنید.
این معرف به دلیل مهار بسیار مؤثر فعالیت RNase ، می تواند یکپارچگی RNA را در طول همگن سازی یا لیز نمونه حفظ کند، در حالی که سلول ها کاملا لیز می شوند .با استفاده از معرف YTzol می توانید از مقدار کمی بافت (50-100 میلی گرم) و سلول ، ریبونوکلئیک اسید (RNA) با غلظت بالا استخراج کنید. همچنین سادگی روش این معرف، امکان پردازش همزمان تعداد زیادی از نمونه ها را فراهم می کند.

دانلود دستورالعمل محلول YTzol Pure RNA
سوالات متداول
در استخراج توسط YTzol تناسب بین اندازه نمونه و مقدار YTzol بسیار حائز اهمیت است.توصیه می شود در این موارد به مقدار مناسب از معرف YTzol برای نمونه خود دست یابید . به طور عمومی مقدار YTzol سه برابر مقدار نمونه اولیه است اما این مقدار بر اساس نوع نمونه قابل تغییر است. بنابراین سعی کنید تا به مقدار متناسب با نمونه خود دست یابید. از آنجایی که در روش استخراج با YTzol مقداری RNA از دست می رود پیشنهاد می شود به دلیل کم بودن مقدار ماده ژنتیکی نمونه و نیز امکان از دست رفتن همان مقدار اندک طی مراحل شستشو ،جهت انجام چنین استخراجی از ترکیب YTzol با کیت ستونی شرکت FAVORGEN استفاده کنید. برخی از نمونه ها به دلیل ماهیت خود دارای فنول می باشند که استخراج با YTzol کار را سخت میکند. توصیه می شود در مشاهده آلودگی فنول تعداد دفعات شستشو با الکل 70 درصد را افزایش دهید. دقت شود ممکن است مقداری از غلظت RNA با افزایش دفعات شستشو کاهش یابد. از طرفی به میزان YTzol اضافه شده و نیز به مدت زمان انکوباسیون نمونه با YTzol نیز توجه شود . در استخراج با روش ستونی به دلیل استفاده از ستون های با کیفیت سیلیکایی و نیز جایگزین شدن بافرهایی با پایه نمک به جهت لیز نمونه با معرف YTzol که مبتنی بر فنول-گوانیدین می باشد، RNA با کیفیت بالاتری حاصل می شود. در واقع چون معرف YTzol pure RNA یک ماده لیزکننده می باشد ؛ بنابراین برای تمامی نمونه ها بکار برده می شود.
